These tumors arise from smooth muscle cells of the myometrium and often produce debilitating symptoms in women. Leiomyomas are poorly understood and mostly ignored tumors; however, they are of great clinical importance due to the progressive uterine and vaginal bleeding associated with their growth. Despite their considerable clinical importance, very little is known about the mechanisms by which these tumors develop and progress. My research interests focus on understanding the fundamental biology of leiomyomas and the nature of tumor progression. Using microarrays and an unbiased systems biology approach, I have identified thousands of differentially expressed genes in human leiomyomas compared to normal myometrium. Importantly, the expression patterns of these genes are associated with myoma pathology. In this application I propose to build on my previous findings to gain further insight into the molecular mechanisms of leiomyoma pathogenesis. Furthermore, I propose to characterize the functional roles of these dysregulated genes in vitro. In the future, I plan to investigate the clinical utility of these new findings and to attempt to develop therapeutic approaches based on the specific biological pathways altered in leiomyoma. Relevance: As many as 40% of women suffer from leiomyoma, a common tumor of the uterus and vagina. In this application, I propose to use an unbiased systems biology approach to identify the mechanisms by which uterine leiomyomas develop and progress.
